Abstract | Tuberculosis (TB) is an infectious illness distressing humanity since ancient times. Every year, the number of cases is rising due to resistance gained by the Mycobacterium tuberculosis (Mtb) strains to the existing antitubercular drugs. They are categorized as first-, second-, or third-line agents based on their discovered and efficacy in therapy. The unregulated use of antitubercular drugs in therapy for years has resulted in multidrug-resistant TB. Here, we discussed the global incidences, pathophysiology of TB and classification, synthetic protocol, mode of action, metabolism, structure–activity relationships, and toxicities of antitubercular drugs. Additionally, a detailed note on drug-resistant TB, solutions to overcome it, and the advances and challenges faced for the discovery of vaccines against drug-resistant TB are discussed. Furthermore, recent advances and the clinical trial status of the antitubercular drugs are summarized. This chapter will help the readers to get a comprehensive overview of antitubercular drugs. |
